how to test for significant phenotype enrichment in phylogenetic tree clusters

I have a phylogenetic tree with strains grouped into specific clusters/lineages. Strains are assigned to phenotypes A and B. How can I test in which clusters strains of phenotype A are significantly enriched? Is a simple chi-square test valid?
