A question about drop-seq and Nextera XT Library prepare
Hi guys!
Recently,I was reading a paper about drop-seq and see the serve provided by illunima(https://www.illumina.com/science/sequencing-method-explorer/kits-and-arrays/drop-seq.html) It said they use the Nextera XT Library Preparation Kit for the cDNA. In my impression , Nextera XT Library Preparation Kit using Tn5 to interrupt the DNA meanwhile adding adapter for sequencing .But the cDNA in drop-seq was made up of PCR primer + cell barcode +UMI +cDNA. I am confused if the cDNA was interrupted, the fragment may not contain cell barcode and UMI,and the position which represent the cell barcode and UMI will be indeterminate.
