I am new to single cell analysis. Can RNA velocity (spliced/unspliced) be calculated solely from the raw file (count matrix) provided in the Supplementary files from GEO?

1. The raw file (count matrix) provided in the Supplementary files from GEO would be spliced data, right?

No, you either have the spliced and unspliced counts available that you need, or you need the fastq files to generate them. Before you dive into analysis or you should read this:
