Mean read length

Dear all,

I have one question. After sequencing I received mean read length distribution 121bp every time. In last two runs I received mean read length distribution 115bp. We are using same workflow, same kits and same sequencing. Could you help me, which parameters and which steps could change read length? From preparing samples to sequencing...

This depends on the technology as well as quality analysis and postprocessing. For Ilumina based systems the read length is specified when setting up the sequencing run.
