Does anyone know of a comprehensive database of synthetic lethality in yeast?

Comprehensive is a relative term, but your best bet is probably to start at SGD. The Boone lab also has a standalone server for genetic interactions called DRYGIN. The BIOGRID is another good resource that aggregates data from multiple papers. It includes physical and genetic interactions for several organisms (a superset of what you're asking for).

The latter two resources should be referenced in the gene description pages in SGD (e.g. GAL4). The "Resources" section in the "Interactions" tab gives you a few other databases to investigate.
