Small Change In Amino Acid Sequence Leading To Distant Change In Conformation?

Hi all,

I'm looking for any example of a protein whose secondary or tertiary structure is significantly changed when a single or small number of mutations occur elsewhere from the affected areas. Significance here does not have to mean loss of function, but rather the protein structure is appreciably altered in some sense.

I have come across a protein of a bacterial phospholipase C that has many examples of prolines in various secondary structural contexts, but I'm looking for an example of one or a few AA changes affecting structure either downstream or globally.

I would appreciate if you could point me in the right direction.

Mutations in C are known to change drastically the protein conformation specially when affects disulfide bonds.
