General Enhancer Questions (Methylation Patterns, ect.)
Hi all,
I have a biological question(s) for ya'll involving enhancers. Feel free to link any relevant studies or review as well.
1.) Enhancers work by being the binding sites for TFs. But how does this increase transcription? (Especially when enhancers can be extremely far away from the gene they control.)
2.) Enhancers can become methylated. When they do, does this usually prevent transcription factor binding? And if it does prevent the binding, how does this affect the role of the TF and gene expression.

Hey Ram, sorry about that! But I'm asking these questions because I'm starting to do some ATAC analysis, and just trying to understand some fundamentals so when I do peak-calling, ect, I know more of what I'm talking about. (the methylation part was more for my understanding however.) Still totally understand if this question is beyond the scope of the site. Cheers!
You should try biology SE/SeqAnswers and while you're there, please avoid statements like "I have a question for y'all" and "Feel free to link any relevant studies or review as well". It sounds condescending, like you're _allowing_ contributors the freedom to help you. Instead, please just ask politely for what you need.
