CRISPR vector redesign

Hi there,

What is the best option for gRNA replacement in a CRISPR vector, considering there is no suitable RE even a few thousands bp around and the vector is 18kb in size. The vector was already constructed through USER cloning, but I'm thinking about other possible alternatives like MOE-PCR.
