Hey dudes,

I downloaded some packages from bioconductor, do I need to re-download them whenever I launch R?

No, you need to loading/attaching them: http://https://stat.ethz.ch/R-manual/R-devel/library/base/html/library.html

Sorry I am downloading biocLite("hugene10stprobeset.db"), it taking long time and I'm still waiting to be finished, do you think if it is normal?

Those probeset packages can be rather large, give it time.
