In my manuscript for publishing, there is a sentence that "ni is the size of NO. 1, 2, 3, …, i unit",

and the reviewer commented that the word "NO." in "No. 1,2,3...." should be expanded to avoid conclusion and increase clarity while reading."

I think I just replace "NO." into "number", but my investigator do not think so. Therefore, how should I correct the expression here.

This is not a bioinformatics question. Probably more appropriate for the English SE. This being said no. is a common abbreviation for number (note though that the abbreviation typically doesn't have a capital O). Without more context, we can't say whether there is a risk of confusion as claimed. Anyway this is not such a big deal, just replace the abbreviation by the full word and be done with it.
