How to characterise a protein "enzyme", and find evolutionary divergence into two distinct pathways?

Hi, I was wondering if any one can tell me, how I can characterise a protein divergence evolution significant in metabolic pathway. I'll be using rDNA technology to express the possible genes of these "proteins" or "enzymes" to be more specific to express them in a vector and then infect them on plants.

I am asking for a short one-liner how to proceed and what parameters should I emphasise the most..?

Any explanation will be of great help.

asking for a short one-liner

I hope you don't mean code?

No, I meant simple steps

like steps represented with ">" or "|"

Example:

Dna_Isolation > Quantification > PCR > Library_preparation ...

This a general review, it may clear some points.
