What do you mean by "extend" the gene family? What kind of data you have?

5 genes are present in one gene family ..i want to extant or add more members in gene family.

..suppose u have a gene name XYZ in one organism. this same gene is present in a closely related organism with the same name as xyz...then how we can differentiate between two sequences.

.then how we can differentiate between two sequences.

What do you mean by differentiate. If a gene has an essential role its sequence may be fully conserved across species and you would not be able to differentiate the two species based on sequence. That said generally critical residues (e.g. active site, contact points for interactions) would be under selection pressure and other amino acids may/will change some over evolutionary time. That is one way you identify orthologs.

thank you so much to clear my concept. my another question is that.one family had 7 members means 7 genes.iwant to add more genes in this family .then what procedure or alignment analysis I have to do?

See the links I provided below to find resources at NCBI/Ensembl.

iwant to add more genes in this family

Just because you want something does not mean that it is scientifically correct or practically feasible. Why do you want to do this? What is the scientific basis/hypothesis you are working with?

orthomcl all the proteins and see if those you know have additional included in orthomcl. google orthomcl and can use web version or download

for the wheat genome, this tool is functional?

I am going to suggest that you take a look at the precomputed sources of data at Ensembl (I am not sure if wheat genome is included). NCBI has book chapters here that will walk you through the process of identifying orthologs..
